Abstract
Adenovirus virions are non-enveloped icosohedra containing a linear double-stranded DNA genome. The viruses are classified into five genera, with one containing the 7 human adenovirus species. Particles enter cells by receptor-mediated endocytosis, escape the endosome and deliver the genome to the nucleus. There, phased gene expression takes place using host RNA polymerase II and the genome is replicated by a virus-coded DNA polymerase. Proteins produced early in infection interact with and adapt the host environment to limit innate and adaptive immune responses so as to favour a productive outcome. Several adenoviruses have been adapted for use as gene delivery vectors.
