Abstract
Paracoccus versutus cytochrome c(552.5) represents one of the major components of a thiosulphate-oxidizing multienzyme system (TOMES). In this investigation, the electrochemical behaviour of cytochrome c(552.5) adsorbed on a glassy carbon electrode has been studied by cyclic voltammetry at pH 7.0, at 20 degrees C. The electron transfer process was found to be irreversible for scan rates above 0.1 V s(-1), with alpha = 1.06, a heterogeneous rate constant of k(s) = 0.45 s(-1), and a standard redox potential of E-0 = + 250 mV (vs. SHE). The redox process was found to involve the transfer of two electrons and a single proton at pH 7.0. Deadsorption of cytochrome c(552.5) from the electrode was investigated by chronocoulometry at pH 7.0 and 200 mV (vs. SCE), and found to be diffusion-controlled. A surface concentration of 2 X 10(-11) mol cytochrome c(552.5) cm(-2) was determined at the start of deadsorption, with an adsorption constant of (beta similar to 10(6) M-1 and a maximum surface concentration of 5 x 10(-11) moles cytochrome c(552.5) cm(-2).
