Abstract
We have developed a non-linear compartmental model incorporating saturable binding to calculate the antibody characteristics and optimal radiation for killing tumours with I-131 and Y-90 labelled antibodies. The model is represented by a series of stiff differential equations which have been solved using the FACSIMILE computer package combined with the SAS graphics package and MIRD radiation dosimetry tables. Results show that a 60 Gray killing dose to the tumour requires a much higher tumour uptake of antibody than presently achieved. Essential antibody characteristics are high affinity combined with the optimal dose for each patient.
