Abstract
Firstly, an overview of current approaches towards the processing of porous biopolymer scaffolds as a template for cell attachment is presented. A comparative parametric analysis of various processing techniques highlighted the importance of scaffold design, pore morphology, interconnectivity and cell infiltration. The author then presents relatively novel methods of fabricating PLLA and PLLA-Bioglass constructs using solvent extraction as well as freeze casting using chloroform, 1,4-dioxane, water and camphene. 3-dimensional scaffolds with high porosity (to a maximum of 90%) and an average pore size of 10-420 µm were produced for polymer and polymer-Bioglass composites. Variation in porosity and the degree of spatial organization of pores changed with polymer/solvent/Bioglass concentration and freezing temperature (a maximum of -196oC). The biocomposite architecture exhibited both macropores (average 200 µm) and micropores (average 10 µm). XRD, DTA-TGA, SEM and micro-CT were used to characterize the poly(L-lactide) based constructs which hold great potential for tissue engineering applications.
