Abstract
Gliomas account for approximately 30% of primary brain tumours and 80% of malignant brain tumours. The clinical hallmarks are characterized as its aggressive growth and inexorable recurrence. A deeper understanding of the genetic and epigenetic alterations occurring in primary brain tumour could help improve the knowledge relevant to new biomarkers discovery. In this project, we have identified a link between coding - genes, lncRNAs and transposable elements that are implicated in the misregulation of genes altered in high - grade glioblastomas by performing an RNA - seq transcriptome analysis. We applied an inducible CRISPR/Cas9 system to edit the genes involved in a potential regulation impacting on the development of glioblastomas. This genome editing approach allows to track the cells for uniform temporal control and efficient gene disruption, and it is also able to target multiple genes simultaneously and decrease off - target effects. According to the intersection analysis , we have found a correlation between PRRX1 and its neighbouring lncRNA, which may impact on the tumour development in glioma. Future work will focus on the investigation of the downstream of cellular pathway that PRRX1 is involved in.
